  • Alternate name: Sutton nevi, Leukoderma acquisitum centrifugum, Perinevoid vitiligo
  • Research fields: Cell biology;Immunology
  • Parental cell: Halo nevus
  • Organism: Human
  • Disease: Halo Nevus
  • Model: Primary line
  • Conditional: No
  • Description: Halo nevus is a mole that is surrounded by a depigmented ring or 'halo'. The formation of a halo surrounding a nevi us believed to occur when CD8+ T lymphocytes destroy the pigment producing melanocytes cells however the cause for the attack is unknown. Halo nevi are estimated to be present in approximately 1% of the general population, and are found to be more prevalent in people with vitiligo, malignant melanoma, or Turner syndrome. Primary melanocytes isolated from a halo nevus can be used as a tool for research.
  • Production details: Autologous skin was taken from halo nevus using a dermatome. Epidermal cell suspension was isolated from the skin sample and seeded in cell culture medium consisting of HAMs F10 supplemented with 10Â?„?žg/ml 12-0-tetradecanoylphorbol 13-acetate (PMA), 0.1nM isobutyl-methyl-1-xanthine (IBMX), 1% Ultroser G, 2mM glutamine, 100 IU/ml penicillin and 100Â?„?žg/ml streptomycin. Overgrowth of fibroblasts and keratinocytes was prevented by addition of geneticine 1/100 (G418).
