  • Organism: Human
  • Tissue: PBMC
  • Growth properties: Impaired Activation
  • Model: Zip6KO
  • Model description: CRISPR/Cas9–expressing Jurkat cell line used to generate a Zip6KO Jurkat cell line
  • Crispr: Yes
  • Description: Cell line used to study how the absence of the Zip6 transporter affects lymphocyte physiology. Zip6 Knockout (KO) line, Zip6 is strongly upregulated early during activiation of T lymphocytes stimulated using anti-CD3/anti-CD28
  • Production details: Cells were maintained in RPMI 1640 medium supplemented with 10% FBS and 1% penicillin/streptomycin. Where indicated, FBS was incubated according to the manufacturer’s instructions with Chelex 100 resin (Bio-Rad Laboratories) to generate Zn2+-free growth medium. Various amounts of ZnSO4 were added to the final medium to generate specific Zn2+ concentration conditions. Cells were grown at 37°C in a humidified 5% CO2 atmosphere.
