  • Tool name: Spag17/Tomato-EGFP mouse
  • Research fields: Cell biology;Organ fibrosis;Primary cilia dyskinesia;Skeletal dysplasia
  • Disease: Development
  • Model: Knock-out
  • Crispr: No
  • Description: This is a flox mouse model for the tramsission of the Spag17 tm1a vector linked to a Tomato-EGFP system for cell lineage tracing. This mouse model has the potential to generate a knockout for Spag17 when crossed with a Cre mouse. Cells will express an EGFP fluorophore for in vivo tracking if they are deficient in Spag17 and presents an avanue for studying primary cili dyskinesia, development, skeletal dysplasia, primary cilia disfunction, aging, infertility, and multi-organ fibrosis.
  • Application: Southern blot, RT-PCR, western blotting, IHC, immunofluoresence, electron microscopy, and magnetic resonance imaging
  • Genetic background: Spag17+/flox mice were mated with CMV-Cre (B6.C-Tg(CMV-cre)1Cgn/J) mice to generate Spag17+/?;CMV-Cre mice. Male and female Spag17+/?;CMV-Cre mice were mated, resulting in Mendelian inheritance for Spag17?/?;CMV-Cre mice demonstrating a mixed background of C57BL/6J–129S4/SvJ
  • Phenotype: Multi-organ fibrosis
  • Zygosity: Homozygous
  • Strain: C57BL/6J wild-type females, 129S4/SvJaeSor-Gt(ROSA)26Sortm1(FLP1)Dym/J, and CMV-Cre (B6.C-Tg(CMV-cre)1Cgn/J) mice
  • Production details: Embryonic stem cells with conditional potential were used to generate chimeric mice which were crossed to C57BL/6J wild-type females. Resulting mice were crossed to 129S4/SvJaeSor-Gt(ROSA)26Sortm1(FLP1)Dym/J mice to remove the Neo cassette. The Spag17+/flox mice were used for mating with CMV-Cre (B6.C-Tg(CMV-cre)1Cgn/J) mice to generate Spag17+/?;CMV-Cre mice. Male and female Spag17+/?;CMV-Cre mice were mated, resulting in Mendelian inheritance for Spag17?/?;CMV-Cre mice. Afterwards, mice dem...
