  • Additional notes: As a backbone for this vector, pROSA26â€�DV2 vector that carries Rosa26 5′â€� and 3′â€�homology arms and enables rapid, modular, and efficient generation of plasmid DNA suitable for homologous recombination into the Rosa26 genomic locus of mouse embryonic stem cells (mESCs) was used (Nyabi et al., 2009, PMID:19279185). This plasmid allows for multisite Gateway recombinationâ€�mediated insertion of three entry DNA sequences which was used here to generate pROSA26â€�GNrep vector, containing a CAGGS synthetic promoter followed by a loxPâ€�dSTOPâ€�loxP cassette, the GNrep cassette, by a selection cassette pAâ€�FRTâ€�PGKâ€�Neoâ€�pAâ€�FRT (linked in "Related" above).
