  • Description: Capan-1 is a human pancreatic ductal adenocarcinoma cell line. These cells grow in adherent tissue culture and display epithelial morphology. In culture, these cells are capable of invading through an extracellular matrix, such as Matrigel. The Capan-1 cells are resistant to 5-fluorouracil, reminiscent of the original tumour from which they were derived. They form poorly-differentiated tumours when injected into immunocompromised mice. These cells harbour a single base-pair deletion in the BRCA2 allele, which results in the expression of a truncated and dysfunctional protein. In addition, they have an oncogenic mutation in K-Ras (G12V) and an inactivating mutation in p53. These cells express elevated levels of the Epidermal Growth Factor Receptor (EGFR) and do not express SMAD4 protein (i.e., SMAD4-null). The Capan-1 cells are useful both as a xenograft model for pancreatic cancer and as a cell system to study the effects of BRCA2-deficiency.
