  • Description: A novel syngeneic Pik3ca+.p53+ /− murine lung cancer cell line (Y856) with orthotopic growth potential, that can serve as a useful preclinical model for evaluating tumour-host interactions, the impact of specific oncogenic alterations on the tumour microenvironment, and immunotherapeutic approaches. This cell line was derived from the primary lung tumour of a R26StopFLP110*fl/+.p53fl/+ female C57BL/6 mouse and is capable of forming orthotopic tumours in immunocompetent C57BL/6 mice. Y856 cells demonstrate genetic recombination at the Pik3ca and Tp53 loci as shown by PCR, and show reduced expression of TP53 and consistent with constitutive PIK3CA activation, show increased pAKT expression without increased total AKT, as shown by western analysis.

  • Production details: A 20 week old R26StopFLP110* fl/+.p53fl/+ female mouse was injected with 2 µl of 10^9 PFU/ml Ad5-CMV-Cre into the left lung. When this animal was euthanized 11 week later, a 6 mm left lung tumour was combined with multiple metastasis (contralateral lung, pericardial, pleural) then passaged into a female recipient animal. After one passage through the flank, the Pik3ca+.p53+/− cell line (Y856) was established; this line was capable of forming orthotopic tumors in immunocompetent C57BL/6 animals.
