Product description: A2780cis is a cisplatin-resistant derivative of the human A2780 ovarian cancer cell line, developed through chronic exposure of the parental cisplatin-sensitive line to increasing cisplatin concentrations. It is used to study platinum resistance, DNA damage response, drug response and cross-resistance mechanisms, and is particularly useful alongside parental A2780 for paired analysis of acquired chemotherapy resistance.

Production details: Split sub-confluent cultures (70-80%) 1:5 to 1:20 i.e. seeding at 1×1,000 to 1×10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C. Cells will attach slowly after resuscitation and take up to 7 days to reach confluency. Recommendation: resuscitate cells in media without cisplatin. Add after subculture of attached cells.
Additional notes: A2780cis is cross-resistant to melphalan, adriamycin and irradiation.

Growth medium: RPMI 1640 + 2mM Glutamine + 1µM cisplatinum + 10% Foetal Bovine Serum (FBS). To retain resistance cisplatin must be added to the media every 2-3 passages.
Subculture routine: Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4 x10,000cells/cm² using 0.05% trypsin or trypsin/EDTA; 5% CO₂; 37°C. Cells will attach slowly after resuscitation and take up to 7 days to reach confluency. Recommendation: resuscitate cells in media without cisplatin. Add after subculture of attached cells.
