Product description: A multiscale germline conditional triple-reporter mouse for in vivo tracking of labelled cells from whole-body to cellular resolution. The model combines murine sodium iodide symporter (mNIS), TagRFP and firefly luciferase (Luc2) in a single Cre-inducible Rosa26 allele, enabling complementary positron emission tomography (PET), fluorescence and bioluminescence imaging in genetically engineered mouse models. This R26LSL-NRL triple-reporter mouse demonstrates robust reporter inducibility across various tissues and enabled the effective tracking and characterisation of tumours in liver and lung cancer mouse models. The model allowed precise pinpointing of tumour location and visualisation of cell-cell interactions within the tumour microenvironment.

Conditional description: Cre-lox conditional reporter system. Cre recombination excises the lox-STOP-lox cassette and activates reporter expression; validated with constitutive CMV-Cre, tamoxifen-inducible R26Cre-ERT2, and tissue-restricted Cre-mediated tumour induction in liver and lung models.

Genetic background: The model was generated by targeting HM1 mouse embryonic stem cells with a Rosa26 construct containing CAG-LSL-mNIS-RFP-P2A-Luc2, followed by blastocyst injection into C57BL/6J mice. Experimental animals were maintained on a mixed 129/C57BL/6J background.
Production details: HM1 mouse embryonic stem cells were electroporated with a SwaI-linearised Rosa26 targeting vector containing the CAGSA promoter, lox-STOP-lox NeoR cassette and mNIS-RFP-P2A-Luc2 cDNA. Correctly targeted clones were identified by PCR across both homology arms, then injected into C57BL/6J blastocysts. Germline offspring were identified by coat colour and genotyped using transgene-specific primers.

R26LSL-NRL conditional triple-reporter mouse. Schematic of the transgene CAG-LSL-mNIS-RFP-P2A-Luc2 in the Rosa26 locus.
