Polo-like kinase (Plx) is a mitotic regulator conserved from yeasts to humans. Many key cell cycle regulators such as p53, cdc25, cyclin B and the Anaphase Promoting Complex are directly targeted by Plx.The Anaphase promoting complex is responsible for the destruction of cyclins and other proteins involved with mitosis.

mRNA encoding either Xenopus ?-globin (x-?globin); Flag-tagged human Aven-WT or Flag-tagged Aven-NES282-292 were added to cycling Xenopus egg extracts to drive protein expression. Aliquots of extracts were then taken at the indicated times and immunoblotted with anti-phospho polo-like kinase-1 (pPlx; upper panels) and antiphospho Tyr15-Cdc2 (Cdc2 pY15; lower panels) antibodies.
