Product description: RPE1 FRT/TR is an engineered RPE1 cell line generated by random integration of an FRT site and Tet repressor gene. Designed for use with the Flp-In™ system, it enables the generation of cell lines with tetracycline-inducible expression of genes of interest, providing a convenient platform for controlled gene expression studies. The line has been used to generate inducible cell models expressing APC15-IRES2-mRuby, APC15-3xFLAG and Cyclin B1-L45A-HA using a modified pcDNA5/FRT/TO vector.

Production details: This tetracycline-inducible RPE1 cell line was created by random integration of an FRT site and a Tet repressor gene into retinal pigment epithelial 1 (RPE1) cells.
Additional notes: Selection markers: This cell line is resistant to Zeocin (FRT cassette) and Blasticidin (Tet repressor cassette). Resistance to Hygromycin and Puromycin is inherited from the parental RPE1 cell line. Flp-In™ integration: The originating laboratory replaced the hygromycin resistance cassette in the pcDNA5/FRT/TO vector with a neomycin resistance marker and used Geneticin (G418 analogue) for selection of successfully integrated clones. Culture recommendations: Use tetracycline-free serum when generating inducible cell lines to minimise background expression from tetracycline-responsive constructs.
