Detection of Sufu; phosphorylated S352 Sufu and HA after immunoprecipitation of HA tagged Sufu Wild Type and Sufu S352A from HEK293T cells. The top panel (pSufu (S352)) was probed with anti-phospho-Sufu polyclonal antibody showing the anti-phospho-Sufu polyclonal antibody is specific and recognises only the phosphorylated form of Sufu but not a mutant where S352 is mutated to alanine. In the middle panel (Sufu); the anti-Sufu polyclonal antibody was used to probe and shows the anti-Sufu polyclonal antibody binds to either form of Sufu independent of the phosphorylation status. The bottom panel shows the same immunoprecipitation preparations probed with an anti-haemagglutinin (HA) antibody; this detects both proteins via their HA tag.
