Sufu is a negative regulator in the hedgehog signaling pathway which directs cell proliferation and patterning during embryogenesis. Sufu down-regulates GLI1- and GLI2-mediated transactivation of target genes and is part of a corepressor complex that acts on DNA-bound GLI1. Required for normal embryonic development. Sufu is ubiquitous in adult tissues. Heterozygous loss of Sufu, in conjunction with the loss of p53, leads to the development of medulloblastoma and rhabdomyosarcoma. Somatic Sufu mutations have been identified in multiple other malignancies, including prostate cancer. |

Detection of Sufu; phosphorylated S352 Sufu and HA after immunoprecipitation of HA tagged Sufu Wild Type and Sufu S352A from HEK293T cells. The top panel (pSufu (S352)) was probed with anti-phospho-Sufu polyclonal antibody showing the anti-phospho-Sufu polyclonal antibody is specific and recognises only the phosphorylated form of Sufu but not a mutant where S352 is mutated to alanine. In the middle panel (Sufu); an anti-Sufu polyclonal antibody was used to probe and shows the anti-Sufu polyclonal antibody binds to either form of Sufu independent of the phosphorylation status. The bottom panel shows the same immunoprecipitation preparations probed with an anti-haemagglutinin (HA) antibody; this detects both proteins via their HA tag.
